Consider below example for your reference.
- User A: Create a new template in “Email Templates” section and add the required signature within the message body.

- Once the template is defined with the signature, you can set this template as Default E-mail template against the individual user by navigating to preferences tab and setting below option from User preferences.
Preference
- Now when this User navigate to Email screen, the default template defined in the preferences settings will be opened with his signature.

Similarly you can define individual User templates with signature and set the Default E-mail template option for each user from preferences tab.
